Local T cell modulation

PLS-Design is using the complexity of the immune system to its advantage. Many failures in pharmaceutical development are based on the fact that a single key element is tinkered with. In this case, the risk is high that either the reaction is not safely controllable, or until then unknown side-ways are activated, rendering the therapeutics ineffective at best.
In contrast, the strategic concept of PLS-Design is to use several switches simultaniously to avoid these pitfalls and gain reliable control over the reaction.

PLS-Design is using a defined local release of active drugs to induce a controlled modulation. The local application is realized by embedding the active substances in an injectable, biodegradable hydrogel beneath the skin. The subcutaneous layer of the skin is populated by many immune competent cells. These modulated cells will migrate and distribute the immune reaction throughout the organism.

The advantage of this system is that a high local concentration of active substances can be maintained for several days without causing adverse side effects, as the systemic application would most likely do. In some cases a systemic application is even unlikely to reach the necessary pharmaceutical concentration at the place of immune reaction.

By local application strategy we are:

  • Minimizing adverse systemic side effects
  • Reducing the needed amount of drug
  • Enabling higher effective doses
